I just replaced my rear DSP33"s with in wall DSP420's. Do I need to create a whole new set up configuration? I couldn't find a fast way to just cut out the DSP33's and replace with the 420's.

Is it even necessary to do this? aren't they essentially the same?

Should be the same I would think. Doubt you need a new profile.
